조직복원술에 적용 가능한 무세포 콜라겐 층의 제조방법에 관한 연구

Abstract
본 논문에서는 외과수술 후 얻게 되는 잉여 피부조직을 이용하여 기존 선행 되어온 무세포 진피기질 제조 방법에서 사용한 SDS 합성계면활성제 등을 대체 한 천연 계면활성제가 잔여 세포들이 세척이 얼마나 잘 되었는지를 관찰하는 것이 연구 목적이다. 기존에 선행되어온 무세포 진피기질 제조방법으로 조직을 염색 후 광학현미경으로 관찰한 결과 NaCl 용액에 담근 후 조직 관찰결과 세포 벽이 없어지고 세척이 되었지만 잔여 세포는 남아 있었다. SDS (Sodium dodecyl sulfate) 용액 처리 후 조직 관찰 결과 잔여 세포들은 완전히 사라지지 않았다. 이에 대체 물질로 첨가물이 들어간 식물성 천연 계면활성제로 사용한 결과, SDS 계면활성제를 사용한 조직과 비교하였을 때 남아있는 잔여 세포가 대부분 없어진 것이 관찰되었다. 제조된 무세포 콜라겐을 솔루션 작업을 통해 액체화 하여 상처 치유분석 방법(Wound Healing Assay Method)을 통해 얼마나 상처 된 부위에 치유가 되는지도 시간적으로 관찰 하였다. 24시간 경과 후 동종 무세포 콜라겐이 Mouse collagen보다 상처 부위가 완전히 치료가 된 것이 관찰되었다.
Alternative Abstract
The purpose of this paper is to observe how well the remaining cells are washed with a natural surfactant replacing the SDS (Sodium dodecyl sulfate), which was used in the existing method of manufacturing the acellular dermal matrix, by using the surplus skin tissue obtained after surgery. The tissue was stained with the existing method for preparing acellular dermal matrix and observed with an optical microscope. Through observation of the tissue after immersing it in NaCl solution, the study found that the cell wall disappeared and was washed but residual cells remained. In addition, treating the tissue with the SDS, did not completely remove the remaining cells. In contrast, the study used natural vegetable surfactant containing additives as an alternative and observed that the remaining cells mostly disappeared compared to the tissue using the SDS. Next, the study liquefied the prepared cell-free collagen through solution work and temporally observed how much the wound was healed through the Wound Healing Assay. After 24 hours, it was observed that allogeneic cell-free collagen healed the wound more than mouse collagen.
