dc.description.alternativeAbstract | The SLC45 family members have been object of study for many years by many researchers. Their structure characteristics similar to each other, they have a role of solute carrier transporter in mammalian. The family is composed of four members called SLC45A1-4, and each of them has a predominant role in specific mammalian tissue. SLC45A1 was initially identified in brain as a sugar transporter. The SLC45A2 is known to participate of protein production that is mediated for melanin synthesis. SLC45A3 is expressed in prostate, and it is associated with prostate cancer. On the other hand, SLC45A4, has been recently hypothesized as a putative transporter in sperm. Despite that, information is still lacking, and further study is still required to precise its localization and function in the mammalian tissue. As a result, in this investigation, the role of Slc45a4 in sperm function and mobility was studied. To investigate whether Slc45a4 is involved in spermatozoa mobility through maturation process, the Slc45a4 gene was silenced in C57BL/6 mice and analyzed to detect the expressions of various genes involved in plant sugar transportation gene metabolism-related and enzymes. Also, Slc45a4 localization in C57BL/6 embryonic day 18.5 mice was analyzed by using in situ hybridization. Furthermore, Slc45a4 deficient mice spermatozoa and reproductive tissue were isolated separately to tissue of male reproductive organ and evaluated by qPCR to observe mRNA expression of glucose and fructose transporters, as well as the metabolism-related enzymes. We demonstrate that the embryonic analyze contribute to the understanding of dominant expression of Slc45a4 in male seminiferous tubule integrated to their transmembrane. Moreover, Slc45a4 present to be a plant sugar transporter for fructose and comport to have similar conservative plant sugar transporter motif. In addition, Slc45a4 knock-out mice use more higher glucose degradation pathway for energy source as compensation for mobility of spermatozoa flagella through the pathway called polyol pathway. It is the pathway which produce fructose from intracellular glucose. In conclusion, the apparent compensation mechanism of Slc45a4 role as fructose transporter by polyol pathway demonstrated that Slc45a4 absence presents no crucial effect on spermatozoa mobility. Also, its deficiency in mice does not affect litter reproduction process for generation after generation. | - |
